TheFeraSpinSeriescomprisessixsuperparamagneticironoxide(SPIO)andundltrasmall(
USPIO)particlesspecificallyformulatedforpre-clinicalmagneticresonanceimaging(MRI):FeraSpinXS,FeraSpinS,FeraSpinM,FeraSpinL,FeraSpinXL,andFeraSpinXXL.Theirwell-definedsizesallowforMRIstudiesthatrelyonspecificparticlesizes.
FeraSpinXStoXXLareagentsofhighrelaxivityandenhancethecontrastinT1-aswellasT2-andT2*-weightedMRI.TheT2effectincreaseswithincreasingparticlesizewhereastheT1effectincreaseswithdecreasingparticlesize.Uponaccumulationincells,theT1effectdiminishesandtheT2effectincreases.
Uponintravenousinjection,allagentsofthisseriescirculateinthebloodstreamandaretakenupbymacrophages.Theyaccumulateintheliverandspleenandaredegradedwithinafewdays.Theironistransferredintothephysiologicalironstores.
Uptakebymacrophagesdependsontheparticlesize.UptakebytheKupffercells(macrophagesoftheliver)increaseswithparticlesize,thusle
ADIngtorapidaccumulationintheliverandspleenandashortbloodcirculationtime.Withdecreasingparticlesize,theuptakebytheKupffercellsisreduced,leadingtoaprolongedcirculationtimeandincreaseduptakebyothermacrophages.
FeraSpinXSshowsthelongestbloodcirculationtimeamongtheFeraSpinproductsandthereforeisparticularlysuitedforvascularstudies.ThisUSPIOparticlehasthehighestT1effectandisspecificallyformulatedformagneticresonanceangiography(MRA).
