D-Val-Lys-Lys-Arg-AMC is used as a fluorogenic substrate for metacaspases. Metacaspases are found in plants, fungi, and protozoa. They are cysteine-proteases and are distantly related to metazoan caspases.
Activity is quantified by release of free fluorescent 7-amino-4-methylcoumarin (AMC) which excites at 360-380 nm and emits at 440-460 nm.
