  TheCLIP-tag?proteinlabelingsystemenablesthespecific,covalentattachmentofvirtuallyanymoleculetoaproteinofinterest.CLIP-tagisaproteintagbasedonhumanO⁶-alkylguanine-DNA-alkyltransferase(hAGT).CLIP-tagsubstratesarederivativesofbenzylcytosine(BC).Inthelabelingreaction,thesubstitutedbenzylgroupofthesubstrateiscovalentlyattachedtothereactivecysteineofCLIP-tagformingastablethioetherbond.AlthoughCLIP-tagisbasedonthesameproteinasSNAP-tag,thebenzylcytosinesubstratesformaseparateclassofsubstrates,differentfromthebenzylcytosinesubstratesformaseparateclassofsubstrates,differentfromthebenzylguaninesubstratesrecognizedbySNAP-tag.CLIP-tagandSNAP-tag?canbeusedfororthogonalandcomplementarylabelingoftwoproteinssimultaneouslyinthesamecells.
